The present study included 202 suspected cases of Poliomyelitis from different areas of Pakistan through expanded programme of immunization (EPI). From each patient two stool specimens were collected 24-48 hours apart and were inoculated on the cell lines derived from rhadomyosarcoma and epidermoid carcinoma. Typing of poliovirus isolates was done by utilizing the neutralization technique. The hospital percentage of suspected cases (38.1%) belonged to 3-12 months age groups, male to female ratio being 1.4:1.0. Among the suspected cases, 94.06% were vaccinated, while 1.98% were partially vaccinated. Poliovirus was isolated from 28.2% stool specimens of suspected cases of poliomyelitis. The poliovirus type 1 was found to be commonest (75.4%) among positive cases. It was concluded that: (I) more than 90% of cases were unvaccinated, and (ii) the most prevalent poliovirus was found to be PI type. It is suggested that: (i) stool cultures for isolation of poliovirus should be performed in every case suspected of poliomyelitis and the finding of positive cases among the vaccinated group suggests the need for observance of proper cold chain maintenance and a change from exclusive oral polio vaccine (OPV) regimen to combined or sequential immunization schedule.
