Molecular detection of high risk human Papilloma viruses in oral squamous cell carcinoma and precancer.

OBJECTIVE: To determine the prevalence of high risk Human Papilloma Virus (HPV) 16/ 18 in oral pre-malignant and malignant lesions using highly sensitive molecular pathology techniques. STUDY DESIGN: Retrospective analysis of paraffin embedded tissue blocks. MATERIAL & METHODS: 130 cases of oral squamous cell carcinoma, 65 cases of oral pre-neoplasia and 65 normal oral mucosal tissues were subjected to high risk HPV 16 & 18 for molecular analysis. RESULTS: Oral squamous cell carcinoma cases displayed 23(17.7%) positive cases for HPV 16/18 by PCR and 19(14.6%) cases showed viral DNA by NISH. The pre-neoplastic lesions revealed viral DNA only in 3 cases and none of the normal oral mucosal tissues exhibited positive signals for HPV. CONCLUSION: HPV was detected in very small number of cases depicting the contributory role of high risk HPVs rather than a necessary step in oral oncogenesis.
