Study of the effect of H2-Receptor Antagonists of Gastrointestinal Motility.

The study wars conducted to see the non-specific actions of climetidine, ranitidine and relatively new compound farmotidine on gastrointestinal motility. The study was done on rabbis ileum and rat fundus strip in vitro. Isotonic contractions were recorded with kymograph. Effects of known agonists like scetyicholine, physostigmine and 5hydroxytryptamine were studied for purpose of comparison. Ranititidine had stimulant effect both on rabbit ileum and rat fundus with ED50 of 32 mcg/ml and 28 mcg/ml respectively. Cimetidine had week stimulant effect on rabbit ileum and no effect on rat fundus strip. Famotidine had virtually no effect on both the tissues. These findings suggest that H2-antagonists have some actions which are unique to individual molecule and data obtained with one antagonist should not be extrapolated to entire class.
