Isolation of heparin and its assay development.

Heparin from lungs of oxen, sheep and buffalo was extracted in crude form by employing Charles and Scott technique with slight modification. Among all, the maximum crude extract was obtained from oxen lungs. An efficient metachromatic method for analysis of heparin was developed. This method involved the measurement of light absorption at 530 nm by a purple complex, formed by toluidine blue with heparin. The crude extract from oxen lung had a titre approximately 75% that of the standard heparin. The bioassay indicated that the results of standard and crude extract samples were approximately in line with metachromatic assay.
