Comparison of different diagnostic methods on skin biopsies in Cutaneous Leishmaniasis.

Comparison of various diagnostic methods was done to detect leishmanial parasites in skin sections of clinically suspected cases of cutaneous leishmaniasis. The skin biopsies were collected mostly from skin center Military Hospital (MH) Rawalpindi. Some of the biopsies received from peripheral hospitals were also included. The methods used were the routine histology using hematoxylin & eosin (H&E) stain, (periodic acid schiff) PAS and Giemsa stains, Indirect fluorescent antibody test (FAT), Peroxidase labeled antibody (PAP) test and k DNA (kinetoplast DNA) probes. The results of these tests were later compared. Peroxidase labeled antibody test and H&E staining had almost the same efficacy. The k DNA probes were tried but technique needs to be perfected before it can be used in routine diagnosis.
