Screening of pathogenic Acanthamoeba specific Antibody fragments using Phage display library.

Genus Acanthamoeba consists of both virulent and avirulent strains / isolates. Clinically, it is important to differentiate pathogenic species of Acanthamoeha from non-pathogens. In this study, different Acanthamoeba isolates were divided into pathogenic or non-pathogenic groups based on cytopathic effects on corneal epithelial cells. Using a phage display library, antibody fragments were isolated which were specific for pathogenic Acanthamoeba. Polymerase chain reaction showed the diversity of these clones. Furthermore enzyme-linked immunosorbent assays and flow cytometry revealed the specificity of two clones only for pathogenic Acanthamoeba. In conclusion, these antibody fragments can be potentially used for the differentiation of pathogenic and non-pathogenic Acanthamoeba.
